This problem can be replicated on the chat portlet. The Chat plugin works properly in the default portal instance, both before and after sharding is set up, but will not successfully send messages in additional instances/shards. To reproduce:
(Before beginning the following steps, you will need to be using a portal-ext file that is properly configured for sharding across multiple databases)
1.) Deploy the Chat plugin
2.) Click Control Panel
3.) Click Portal Instances
4.) Create a new instance
5.) Log in to the new portal instance
6.) Create a new user
7.) Open a different browser
8.) Log in as the new user on the new portal instance
9.) Send test messages from one user to the other
This issue was tested in the following environments:

Tomcat 6.0 + MySQL 5. Firefox 3.6.17. 6.0.x Revision 80400.
Tomcat 6.0 + MySQL 5. Firefox 3.6.17. 6.1.x Revision 80400.
